Mechanism of action

Melphalan hydrochloride alkylates DNA, forming cross-links and inhibiting DNA replication and RNA transcription. This disrupts cellular function and leads to apoptosis in rapidly dividing cells, particularly in malignant cells.

Indications

Multiple myeloma, ovarian cancer, neuroblastoma, rhabdomyosarcoma, breast cancer, malignant melanoma, soft tissue sarcoma.

Side effects

Myelosuppression, nausea, vomiting, diarrhea, mucositis, alopecia, hepatotoxicity, pulmonary fibrosis, secondary malignancies, severe hypersensitivity reactions, cardiac toxicity.

Precautions

Contraindications

Hypersensitivity to melphalan or excipients, lactation, severe bone marrow suppression, pregnancy, recent vaccination with live vaccines, severe renal impairment.
