Dose

Adults: 1–5 mg orally daily. Pediatrics: For premature labor, 5 mg orally every 6 hours; for prostate cancer, dosing individualized. Use in pregnancy is contraindicated. Mechanism of action

Binds to and activates estrogen receptors (ERα and ERβ), modulating gene transcription; this leads to increased synthesis of estrogen-responsive proteins, resulting in proliferation and differentiation of target tissues, inhibition of gonadotropin secretion via negative feedback on the hypothalamic-pituitary-gonadal axis, and altered reproductive tract development.
