Sustained-release capsule

API strength per dosage

Domperidone 30 mg, Esomeprazole 40 mg per capsule.

Dose

Adults: One capsule once daily, 15–30 minutes before a meal. Not recommended for children under 12 years or weighing less than 35 kg. Use with caution in elderly or those with hepatic or renal impairment; dose adjustment may be required. Avoid in patients with moderate to severe hepatic impairment.

Mechanism of action

Domperidone antagonizes dopamine D2 receptors (DRD2) in the chemoreceptor trigger zone and gastrointestinal tract, enhancing acetylcholine release and promoting gastric motility. Esomeprazole irreversibly inhibits the H+/K+-ATPase enzyme (proton pump) in gastric parietal cells, suppressing gastric acid secretion.

Precautions

Contraindications

Hypersensitivity to domperidone or esomeprazole, prolactin-releasing pituitary tumor, gastrointestinal hemorrhage or perforation, severe hepatic impairment, concomitant QT-prolonging drugs, known cardiac arrhythmias, pregnancy, lactation.
