Drug classification

Dosage form

Film-coated oral tablet containing 90 mg of etoricoxib.

Dose

Etoricoxib 90 mg is typically used once daily for adults, with dose and duration depending on indication. Not recommended for children or adolescents. Use lowest effective dose for shortest duration. Contraindicated in severe hepatic impairment; use with caution in mild-to-moderate hepatic or renal impairment. Always consult local prescribing information for specific recommendations.

Mechanism of action

Etoricoxib selectively inhibits cyclooxygenase-2 (COX-2), reducing prostaglandin synthesis involved in pain and inflammation. It has minimal effect on COX-1 at therapeutic doses, thereby sparing gastric mucosa and platelet function. The result is decreased inflammation and analgesia with a lower risk of gastrointestinal side effects compared to non-selective NSAIDs.

Pregnancy category

Etoricoxib should be avoided in pregnancy, especially in the third trimester, due to potential risks of fetal harm and adverse effects on fetal circulation.

Precautions

History of cardiovascular disease, hypertension, renal or hepatic impairment, gastrointestinal bleeding or ulcer risk, elderly patients, concurrent anticoagulant or antiplatelet use, monitor blood pressure, renal and liver function, signs of GI bleeding, avoid in pregnancy and lactation, use lowest effective dose for shortest duration.

Contraindications

Hypersensitivity to etoricoxib, active peptic ulcer or gastrointestinal bleeding, severe hepatic impairment, severe renal impairment, inflammatory bowel disease, congestive heart failure (NYHA II-IV), uncontrolled hypertension, pregnancy, established ischemic heart disease, peripheral arterial disease, cerebrovascular disease.
