Oral antidiabetic; sulfonylurea (glimepiride) and biguanide (metformin) combination.
Film-coated oral tablet containing glimepiride and metformin hydrochloride.
Glyree-M 1 Tablet PR combines glimepiride 1 mg and metformin 500 mg, typically taken once daily with meals. Dose adjustments may be needed based on glycemic response. Not recommended in severe renal or hepatic impairment. Safety and efficacy in pediatric patients are not established. Always individualize dosing and monitor renal function regularly.
Glimepiride stimulates insulin release by binding to pancreatic β-cell sulfonylurea receptor 1 (SUR1), closing ATP-sensitive potassium channels. Metformin decreases hepatic glucose production and increases insulin sensitivity by activating AMP-activated protein kinase (AMPK). Together, they lower blood glucose through enhanced insulin secretion and improved peripheral glucose uptake.

Glimepiride and metformin should generally be avoided in pregnancy; insulin is preferred for glycemic control due to limited safety data and potential fetal risks.
Monitor blood glucose, renal function, liver function, signs of hypoglycemia, gastrointestinal symptoms, vitamin B12 levels, avoid in severe renal/hepatic impairment, caution in elderly, assess for lactic acidosis risk, avoid excessive alcohol, review for drug interactions, monitor weight, counsel on diet and exercise adherence.
Hypersensitivity to glimepiride, metformin, or sulfonylureas; diabetic ketoacidosis; severe renal impairment; acute or chronic metabolic acidosis; severe hepatic impairment; conditions predisposing to hypoxia; alcoholism.
