Mechanism of action

Ofloxacin inhibits bacterial DNA gyrase and topoisomerase IV, enzymes essential for DNA replication, transcription, repair, and recombination. This inhibition prevents supercoiling and separation of bacterial DNA, leading to impaired cell division and ultimately bacterial cell death.

Pregnancy category

Precautions

Assess for hypersensitivity, history of tendon disorders, CNS disorders (seizures), renal impairment, risk of QT prolongation, photosensitivity, pediatric use caution, monitor for tendon pain/swelling, CNS effects, allergic reactions, liver and renal function, avoid with antacids or iron/zinc within 2 hours, ensure adequate hydration.

Contraindications

Hypersensitivity to ofloxacin or other quinolones, history of tendon disorders related to quinolone use, epilepsy, history of QT interval prolongation, children or adolescents during growth, pregnancy, breastfeeding.
