Adults: 500 mg every 6 hours. Children 2–10 years: 250 mg every 6 hours. Children <2 years: 125 mg every 6 hours. Renal impairment: No routine dose adjustment; monitor for accumulation in severe impairment. Administer 30–60 minutes before meals.
Flucloxacillin inhibits bacterial cell wall synthesis by binding to and inactivating penicillin-binding proteins (PBPs), particularly transpeptidases, preventing cross-linking of peptidoglycan chains. This weakens the cell wall, leading to osmotic instability and bacterial cell lysis. It is resistant to degradation by staphylococcal beta-lactamases.

History of allergy to penicillins or beta-lactams, hepatic dysfunction, renal impairment, monitor liver and renal function, monitor for hypersensitivity reactions, monitor for signs of superinfection, caution in elderly, monitor for gastrointestinal disturbances, assess for drug interactions, use in pregnancy and lactation only if clearly needed.
Hypersensitivity to flucloxacillin, other beta-lactam antibiotics (e.g., penicillins, cephalosporins), history of flucloxacillin-associated jaundice or hepatic dysfunction, serious allergic reactions to penicillins.
