Adults: Acute gout flare, 1.2 mg at first sign of flare followed by 0.6 mg one hour later; maximum 1.8 mg over one hour. Prophylaxis, 0.6 mg once or twice daily. Pediatrics: Not recommended for children under 4 years. Adjust dose in renal or hepatic impairment.
Mechanism of action
Colchicine binds to tubulin, inhibiting its polymerization into microtubules, thereby disrupting cellular functions such as mitosis and neutrophil motility and activity, leading to reduced inflammation and pain in conditions like gout.
