Mechanism of action

Binds to the α2δ-1 subunit of voltage-gated calcium channels (Cavα2δ-1) in the central nervous system, inhibiting presynaptic calcium influx, thereby reducing release of excitatory neurotransmitters such as glutamate, noradrenaline, and substance P, resulting in decreased neuronal excitability and analgesic or anticonvulsant effects.

Indications

Partial seizures (with or without secondary generalization) in adults and children aged 6 years and above, peripheral neuropathic pain such as diabetic neuropathy and postherpetic neuralgia in adults.

Precautions

Renal impairment, history of substance abuse, suicidal ideation, abrupt discontinuation, elderly patients, pregnancy, lactation, CNS depression, concomitant use with opioids, driving or operating machinery, respiratory depression risk, hypersensitivity reactions, monitoring for mood or behavioral changes.

Contraindications

Hypersensitivity to gabapentin or any excipients, severe renal impairment without dose adjustment, hereditary galactose intolerance, Lapp lactase deficiency, glucose-galactose malabsorption.
