For erectile dysfunction in adults, the recommended dose of Tadalafil is 10 mg taken prior to anticipated sexual activity, with adjustments to 20 mg or reduced to 5 mg based on efficacy and tolerability. Not recommended for use in pediatrics. Adjust dose in patients with severe renal impairment.
Tadalafil inhibits phosphodiesterase type 5 (PDE5), an enzyme that degrades cyclic guanosine monophosphate (cGMP) in the corpus cavernosum. This inhibition enhances the effects of nitric oxide-induced cGMP, leading to increased blood flow and facilitating penile erection.
Erectile dysfunction, benign prostatic hyperplasia, pulmonary arterial hypertension.
Avoid use with nitrates, severe cardiovascular disorders, recent stroke or myocardial infarction, severe hepatic impairment, hypotension, renal impairment, predisposition to priapism, hereditary degenerative retinal disorders, concurrent use of alpha-blockers or antihypertensives, caution in peptic ulcer disease, anatomical penile deformation.
Hypersensitivity to tadalafil, concurrent nitrate therapy, severe cardiac conditions, recent myocardial infarction, stroke, unstable angina, uncontrolled hypertension, severe hepatic impairment, loss of vision due to NAION.
