OLANZEPINE 10MG

Dose

Adults: 10 mg by intramuscular injection as a single dose; may repeat every 2 hours if needed, up to 30 mg/day. Not recommended for pediatric patients. Use caution in elderly or hepatic impairment; consider lower initial dose (5 mg). Avoid in severe hepatic impairment.

Mechanism of action

Antagonizes serotonin 5-HT2A/2C, dopamine D1–D4, histamine H1, muscarinic M1–M5, and adrenergic α1 receptors, reducing dopaminergic and serotonergic neurotransmission, thereby modulating mood, cognition, and psychotic symptoms.

Indications

Acute agitation associated with schizophrenia, acute agitation associated with bipolar I mania.

Precautions

Monitor for post-injection sedation and delirium, avoid in patients with known hypersensitivity, use caution in elderly or those with dementia-related psychosis, monitor for orthostatic hypotension, avoid concomitant CNS depressants, assess for risk of neuroleptic malignant syndrome, monitor glucose and lipid levels, use caution in hepatic impairment.
