Lineweaver-Burk Plot
Start with the big picture
The plot graphs 1/V₀ against 1/[S], producing a line whose y-intercept is 1/Vmax, x-intercept is −1/Km, and slope is Km/Vmax. These features connect the graph to the enzyme’s kinetic parameters. Inhibitors produce characteristic changes in line position and intercepts: competitive inhibition changes the slope and x-intercept while leaving the y-intercept unchanged; non-competitive inhibition changes the slope and y-intercept while the x-intercept remains unchanged; uncompetitive inhibition produces parallel lines; and mixed inhibition changes both intercepts and the slope. Interpretation has limits: extrapolation near intercepts can magnify experimental error, and the method relies on assumptions such as steady-state kinetics and a single substrate. Alternative linear plots are also considered.
